Research Abstract

ナメクジの嗅覚中枢である脳神経節前脳には多数の神経細胞が存在し,これらが自発的に膜電位の同期振動を生じている.この自発活動は脳神経節の単離後も長時間安定して持続することから,さまざまな測定法のスクリーニングに適した実験系である.これまでの研究により,市販の膜電位感受性色素di-4-ANEPPSによって前脳全体で同期した周期的蛍光変化が記録されることが明らかになった.この蛍光変化は1%程度であり,生理的状態の神経細胞が示す蛍光変化としては極めて大きく,また安定して長時間記録できることから,本実験系は膜電位感受性色素のスクリーニングに適していることがわかる.リポソーム-バリノマイシン系による予備実験で膜電位依存性を持つことが示されたクマリン系およびロドシアニン系の色素を用いて脳神経節を染色し,一次スクリーニングとして染色の状態と自発的蛍光変化の有無を調べた結果,クマリン系色素が強く脳組織を染色することが示されたが,前脳の膜電位振動に伴う蛍光変化を示す色素は存在しなかった.これは用いたクマリン系色素が極性基を持たず,細胞膜内での配向が一定しないためではないかと考え,これにスルホン酸基を付加した色素を合成した.これを用いて脳神経節を染色すると,脳組織は同様に強く染色されたが,やはり膜電位変化を反映した蛍光変化は得られなかった.本研究の結果,ナメクジの単離脳標本が膜電位感受性色素のスクリーニングに有用であること,膜電位感受性色素として機能するためには少なくとも色素の細胞膜への移行と,細胞膜内での配向という2つの要因が重要であること,またリポソームの実験で膜電位応答性のある色素が必ずしも生体で膜電位応答性を示とは限らず,生体を用いたスクリーニングが必要であることが明らかになった.
