Research Abstract

これまでにインテグリンを介する細胞の接着と運動制御において、paxillinとp130Casのチロシンリン酸化が重要な役割を果たすこと、その内paxillinのTyr31/118のリン酸化が上皮細胞の運動先端部におけるRhoAの活性抑制に関与してしていることを明らかにしてきた。今回、paxillinのTyr40/181のリン酸化が、運動する上皮細胞においてCdc42とRac1の活性制御に深く関与することを明らかにし、そのことによりpaxillinが細胞運動極性の制御に深く関わることを示した。即ち、これまでの我々の知見(JBC,275,27155(2000))に加え、今回FRET法(阪大、微研、松田道行先生との共同研究)での評価も行った結果、Tyr40/181のリン酸化変異体を発現させた上皮細胞では、運動中にCdc42とRac1の正常を逸脱した活性化が起っていることを明らかにすることができた異常な活性化が起る部位とその方向への運動極性のぶれは概ね一致していた。Tyr31/118の変異体ではこのようなことは観察されなかった。この異常な活性化は細胞の局所でのみ観察されることであり、細胞全体のCdc42やRaclの活性量を生化学的に測定しても、コントロールと比べ有為な変化を補足することはできなかった。現在、Cdc42やRac1の活性制御に到るTyr40/181のリン酸化の下流シグナル分子を同定する為、最近、Birchmeirらが開発した新しいTwo hybrid systemを導入し、解析を進めている。
