Research Abstract

本研究は糖鎖認識分子であるレクチンのライブラリーをバイオインフォーマテイックスの道具として用いて細胞の分別同定を行おうとするものであり、極めて独創的なものである。特に、レクチンを固定化したものに対する細胞の結合性によって細胞の特徴を明らかにし、個々の細胞を同定できれば、細胞治療に使用するための細胞の同一性の判定基準として極めて有用であると考えられる。本年度には、Maackia amurennsis hemagglutininのループCの131及び133番目のアミノ酸を遺伝子的にランダムに改変した400個のレクチンのライブラリーからランダムにクローニングしたもの52種類(ライブラリー1)と、同じレクチンのループDの糖鎖認識部位の間(218と219、219と220、220と221、221と222、222と223、及び223と224の間)に一アミノ酸を挿入したもの合計120種類の全てをクローニングしたもの(ライブラリー2)、及び以前より得ていたループC内の6個のアミノ酸をランダムに改変したものを含むファージライブラリーからヒト赤血球に対して強い結合性を有するものを選別して得たもの10種(ライブラリー3)、を用いた。これらにFLUGタグをリンクして大腸菌に発現させ、簡便に精製する方法を確立した。一方これらの改変レクチン蛋白質を1ステップで固相化することに成功し、この表面を用いて細胞とレクチンとの相互作用を比較することを可能にした。これらの固相化した複数の改変レクチンを用いて、糖鎖認識特異性の解析を行ない、それぞれが異なる特異性を有することを明らかにした。レクチンと細胞との相互作用がこの固相化した状態で比較した。今回は、肺への転移性を異にする2種類のメラノーマ細胞(マウスB16-F1とB16-F10細胞)、肝臓への転移性を異にする2種類の大腸癌細胞(マウスcolon38及びSL4細胞)への、これらのレクチンライブラリーの結合性を比較することによって、この測定系の信頼性を検証した。ライブラリー1に対しては糖鎖結合性を持つものが多く含まれるが、これらの細胞は何れも結合性が無かった。ライブラリー2に対しては4種類の細胞に結合するものが10種類あり、何れもcolon38の結合性がSL4細胞よりも高いが、B16メラノーマ細胞バリアント間では結合性に違いがなかった。グループ3の全てのレクチンに対してB16メラノーマ細胞(F1及びF10)は高い結合性を示したが、colon38細胞及びSL4細胞は10種類中の2種類に対しては全く結合しなかった。以上のように遺伝子改変植物レクチンをライブラリーとして用いると細胞の分別同定が可能になることを、本研究によって明らかにした。
