Outline of Annual Research Achievements

Eukaryotic translation initiation factor 4E (eIF4E) は、転写後の標的遺伝子産物のタンパク質発現量を制御している翻訳制御の一つである。eIF4Eは、ヒト悪性腫瘍の30%において発現増強していることが知られている。一方、GANPは哺乳類細胞においてRNA輸送に関与する分子で、eIF4Eと細胞質内で結合することを見出している。GANPもeIF4Eと同様に、臨床標本を用いた研究からホジキンリンパ腫、急性骨髄性白血病、慢性リンパ性白血病などさまざまな血液疾患において発現増強が認められている(Fujimura et al, Cancer Res. 2005; Kageshita et al, J. Dermatol. Sci, 2006)。B細胞特異的にGANPを高発現させたトランスジェニックマウスでは、ホジキン様のB-lineage悪性リンパ腫を発症した。さらに、GANP発現上昇が悪性黒色腫や胆管細胞癌と関連していることが報告されている。しかしながら、哺乳類細胞におけるGANPの機能とがん発症に関する分子機構については、まだ明らかではない。これまでの予備実験から、eIF4Eのリン酸化活性が、内在性GANPの発現抑制によって変動することを見出した。この知見は、発がん誘導と密接に関連していることを強く示唆し、本研究計画ではこの分子機構に焦点をあてて解明に取り組むことを目的とする。平成26年度は、ヒトB細胞におけるmRNA翻訳制御におけるGANPの関与について取り組み、GANPがmRNAの5’末端にm7Gキャップ構造を有するタンパク質群と相互作用することを確認することができた。また、標的mRNAについては、GANPがc-Myc mRNAと結合することからc-mycの翻訳活性化にキャップ構造結合要素eIF4EとGANPとの密接な関連性が示唆された。
