Research Abstract

アクチン細胞骨格の再構築は細胞の形態変化、移動、分裂などにおいて重要な役割を果たしている。本研究では、LIMキナーゼ(LIMK)とSlingshot(SSH)によるコフィリンのリン酸化、脱リン酸化経路を中心に、細胞骨格を制御するシグナル伝達機構の解明と、細胞運動、形態変化を時間的、空間的に統御するシステムの解明を目的に研究を行い、以下の結果を得た。
1)ケモカインによるT細胞の遊走におけるコフィリンのリン酸化の時間的、空間的制御の重要性を解明した。細胞遊走因子SDF1によるJurkat細胞の刺激において、コフィリンの一過的なリン酸化の誘導、SSH1とコフィリンの先導端への集積を明らかにした。また、LIMK1、コフィリンの発現抑制は細胞運動、遊走を抑制したが、SSH1の発現抑制は遊走のみを抑制した。LIMK1、SSH1、コフィリンのsiRNA細胞のタイムラプス観察の結果、LIMK1はラメリポディア形成能により,コフィリンはアクチン再編成能により、細胞運動に必須であり、SSH1は膜突起を一方向に制限することにより、細胞の極性形成と方向性のある運動(遊走)に必須であることを解明した。
2)LIMK1を活性化する新たなシグナル経路を解明した。血管新生因子であるVEGFによる血管内皮細胞のアクチン骨格再編成、運動能促進において、LIMK1の活性化が必要であることを示した。また、p38MAPキナーゼの下流で活性化されるMAPKAPK-2によりLIMK1がリン酸化,活性化されることを見出した。リン酸化部位を変異したLIMK1はVEGFによる血管内皮細胞の運動と管腔形成を阻害することを見出し,本シグナル経路の重要性を解明した。
3)SSH1はFアクチンとの結合によって活性化されることを報告したが、今年度はSSH1分子内の3カ所のFアクチン結合部位を同定し、活性化に必要な領域を同定した。
