Outline of Annual Research Achievements

本研究では赤外分光法を用いて、紫外線によって損傷したDNA((6-4)光産物)を修復する(6-4)光回復酵素を対象に、(6-4)光回復酵素が機能を発現するための活性化における構造変化及び光修復における反応メカニズムを解明し、活性化に必要な構造変化が何であるのか、光修復の過程でどのような化学反応が起こっているのかを明らかにすることを目的とする。本年度の研究成果の概要を以下に記述する。
1.(6-4)光回復酵素の各酸化還元状態における基質結合の影響を明らかにするため、各酸化還元状態における基質と結合した構造情報を得るために、活性化過程における赤外差スペクトルを基質がある場合とない場合で比較した。その結果、光活性化過程における基質のC=O基を帰属することに成功し、(6-4)光回復酵素が還元型である時の基質の構造情報を決定したこの研究結果は2016年にBiochemistry誌にて発表した。
2.(6-4)光回復酵素の(6-4)光産物修復に必須と考えられていたヒスチジン残基を変異しても、光照射時間を延ばすことで、(6-4)光産物が修復されることを見出した。これにより、必須と考えられていたヒスチジン残基がなくても酵素は修復機能をもつこと、ヒスチジン残基は反応性を高めるためにはたらくことが明らかになった。この研究結果は2015年にBiophys. Physicobiol.誌にて発表した。
3. 修復中間体を検出するために低温(77 K~)での測定を試みたところ、(6-4)光産物の修復中間体に由来すると考えられる信号を含む3つのスペクトル成分(77, 200, 230 K)を捉えることに成功した。基質の同位体標識した基質試料を用いた測定を行ったところ、電子は5’側の基質に移ること、OH基の転移反応は200 Kで起きている新たな修復モデルを提唱する結果を得た。
