Outline of Annual Research Achievements

平成26年度は、薬物の毒性評価系に応用できる高機能な（高い薬物代謝能を有する）ヒトembryonic stem (ES)/induced pluripotent stem (iPS)細胞由来肝細胞を作製するために、肝細胞分化誘導技術のさらなる改良を行った。肝細胞の前駆細胞である肝幹前駆細胞をラミニン111上で維持培養することにより、肝幹前駆細胞を純化できることを見出した。このように純化して調製した肝幹前駆細胞を肝細胞へ分化誘導したところ、ヒトiPS細胞株に依らず、高効率にアルブミンやアシアロ糖タンパク受容体1を発現する肝細胞を作製できた。したがって、本分化誘導法を用いることによって、ヒトiPS細胞株に依らず高効率に肝細胞を作製可能であることが示唆された。
次に、ヒトES/iPS細胞由来肝細胞を創薬応用できる可能性を検証するため、その薬物代謝酵素cytochrome P450（CYP）をはじめとした各種肝機能をヒト初代培養肝細胞と比較した。ヒトES/iPS細胞由来肝細胞におけるCYP1A2、CYP2C9、CYP3A4活性はヒト初代培養肝細胞と比較して約40-60%程度であった。さらに、ヒトES/iPS細胞由来肝細胞が薬物の毒性試験に応用できるかどうか検討した。ヒトES/iPS細胞由来肝細胞に対して肝毒性を生じることが報告されているベンゾブロマロン等の薬物を作用させたところ、ヒト初代培養肝細胞に近い感度でこれらの薬物の毒性を検出可能であった。以上のことから、本研究で確立した分化誘導法を用いて作製したヒトES/iPS細胞由来肝細胞が創薬スクリーニングに応用できる可能性が示唆された。
