Outline of Annual Research Achievements

本研究は、植物の生育に必須であるリンに対する植物応答分子機構を理解し、低施肥農業への応用を目指した基礎的知見を得ることを目的としている。環境中のリンの増加に伴い植物内での遺伝子発現が上昇するBT遺伝子(BTB and TAZ domain protein遺伝子)familyについて解析した。BT1、BT2遺伝子の発現量はリン投与後、30分後に7-10倍に増加し、6時間以内には3-5倍に安定することが示された。また、タンパク質合成阻害剤による発現誘導への影響について調べた結果、CHX処理を行った場合でも発現誘導は生じており、発現誘導について新規のタンパク質の合成は必要ない応答であることが分かった。一方、CHX処理により遺伝子発現上昇とその後の減少のパターンが見られなかった。このことから、何らかの新規合成タンパク質による発現産物の負の制御機構が存在することが考えられた。さらに、これらの遺伝子発現制御機構とPHR1/PHL1（リン欠乏応答機構の中心的な転写因子）との関係を調べた結果、低リン環境においてphr1/phl1変異体株でのBT遺伝子発現量が野生型株より高く保たれていた。このことから低リン時の発現量減少は、PHR1/PHL1制御下で負の制御を受けることがしめされた。タンパク質合成阻害剤処理の実験から、PHR1/PHL1直接ではなくPHR1/PHL1により誘導されるタンパク質の合成が必要な応答であることが分かった。植物体のリン量は、BT遺伝子欠損株が野生型株よりも多く、放射性同位体元素（P-33）によるリン酸の移行分布は、変異体株のShoot/Root比が野生型株よりも2-4倍ほどが高かった。BT2遺伝子の発現は維管束周辺に見られたことからも根と地上部間の輸送に関する何らかの機能を持つ可能性が示された。
