物理的刺激による多能性幹細胞の精密分化制御法に関する基礎的研究

Outline of Annual Research Achievements

再生医療の実用化において,効率が良く,コストが低い分化誘導法の確立は不可欠である.従来手法では複数の分化誘導因子を組み合わせて用いるが,効率およびコストの両面から改善が求められている.以上の背景を基に,電気刺激などの物理的な刺激による分化制御法の確立を目指すのが本研究の立場である.計画最終年度にあたる本年度は,未分化状態のiPS細胞の電気刺激実験,およびヒトiPS細胞由来神経細胞の成熟過程の評価を行った.
基板上でヒトiPS細胞を培養し,電気刺激を印加したところ,刺激に対する短期的応答として細胞内Ca2+濃度の上昇が起こることを発見した.さらに,細胞外液を変化させた実験,および薬理実験より,電気刺激によるiPS細胞の細胞内Ca2+濃度の上昇は,刺激がトリガーとなり,細胞外のCa2+が細胞内に流入し誘発されること,およびT型Ca2+チャネルは関与しないことが示された.次に,3日間持続的に刺激を続けることにより,ヒトiPS細胞の遺伝子発現量の変化が起こることを示す結果が得られた.以上から,電気刺激が未分化状態のヒトiPS細胞の分化方向に影響を与えることが示唆された.
分化誘導した細胞が神経細胞としての機能を獲得したことを確認するため,微小電極アレイ法による活動計測を行った.分化誘導を行ってから50日目から,10日ごとに計測を行った.結果,ヒトiPS細胞由来神経細胞が分化誘導操作を行ってから,およそ90日後から神経回路網を形成したことが示された.さらに,活動パターンの変化は300日目程度まで持続することが示された.
