Outline of Annual Research Achievements

フグ毒テトロドトキシン (TTX) は強力な神経毒であり、世界中で食中毒事例が報告される。複雑な構造を有するTTXの生合成経路は未だ解明されておらず、起源生物の遺伝子解析から生合成経路に言及した研究もない。昨年度、特徴的な骨格構造を有するTTX類縁体を発見し、その構造からモノテルペンを出発物質とする生合成経路を提唱した。本年度は推定した経路を裏付けることを目的とし、推定生合成経路における中間体を質量分析器を用いて網羅的に探索した。検出された5つの候補化合物を単離し、NMRを用いて構造解析したところ、いずれも含グアニジン六員環を有する二環性の新規化合物だと同定された。得られた化合物群は推定した経路を支持する構造を有していた。これらの成分は複数の有毒イモリ (TTXを有するイモリ) に共通して存在する一方で、無毒のイモリからは検出されず、本化合物群がTTXの関連物だと示唆された。本研究結果は我々が唱えたモノテルペン説を支持しており、陸上生物におけるTTXの生合成経路解明への重要な知見を与えたと考えている。本内容の投稿論文が受理され、公表予定である。
昨年度までに研究室で飼育したイモリに毒の生産能がないことを確かめ、本年度に投稿論文として公表した。本年度は得られた無毒のイモリを用いて、毒の蓄積能を調査した。TTXおよびその類縁体をそれぞれ無毒イモリに経口投与し、一か月後、皮や筋肉を含む身体組織と腹腔内の内臓に分け、LC-MSで解析した。その結果、主に身体組織からTTX、その類縁体が検出され、イモリが経口投与された毒を身体組織に蓄えることが明らかになった。野生のイモリにおいても皮に多く毒が含まれることが知られており矛盾しない。飼育イモリが毒の生産能を示さないことと併せて考えると、イモリの毒は外因性である可能性が高いといえる。長年議論されていたイモリにおけるTTXの起源解明に近づいた。
