Outline of Annual Research Achievements

1)亜鉛超集積植物Noccaea japonica由来ZNT1/ZIP4のsplicing isoformの機能解析: 本年度は、アブラナ科植物が共通して持つZNT1/ZIP4の生理学的機能を明らかにすることを目的に、シロイヌナズナのAtZIP4の機能解析を行った。ノックアウト変異株と過剰発現株を用いた解析の結果、AtZIP4はZn輸送体をコードしないことが示唆された。一方、N. japonicaが獲得したsplicing isoformと同様、N末端に欠損を持つAtZIP4の人工ORFをシロイヌナズナに過剰発現させたところ、植物中のZn濃度は有意に上昇し、N末端欠損型のAtZIP4がZn輸送体をコードすることがわかった。これまでの研究結果から、アブラナ科が共通して持つZNT1/ZIP4の機能は不明であるが、N. japonicaを含めたZn超集積性Noccaea属植物は、高重金属土壌環境に適応する過程でZn輸送体をコードするZNT1/ZIP4のsplicing isoformを獲得したと考えられた。

2)栄養欠乏ストレス下における選択的スプライシングの網羅解析: 本年度では、前年度までに絞りこまれた候補遺伝子の中から、K欠乏に特異的に応答してsplicingパターンが変化する転写因子遺伝子MYB59に着目した。RNA-Seqおよび定量PCRによる発現解析の結果、MYB59遺伝子からは主にDNA結合モチーフをコードする領域を一部欠損したisoformが転写されるが、K欠乏下では完全なDNA結合モチーフをコードするisoformの割合が有意に増加することが明らかとなった。先行研究において、MYB59の発現は主根の伸長を抑制する役割があることが示されている。本研究結果は、MYB遺伝子のsplicingパターンの変化を介した、栄養欠乏に応答した根の伸長制御の存在を示唆している。
