Outline of Annual Research Achievements

昆虫に便乗（乗り物として利用）する線虫Caenorhabditis japonicaは、宿主探索時に重力認識行動を示し、昆虫付着時に長期生存する。本研究では、線虫の重力認識機構および長期生存機構の解明を目的としている。
平成25、26年度は、線虫C. japonicaの重力認識時、昆虫付着時、の生理状態を明らかにするために、RNAseq法およびRNAi法を用いて、それぞれの状態で発現している遺伝子の同定および機能解析を試みた。しかし、C.japonicaは雌雄異体であり、データベースのゲノムの質も悪いため、これらの解析に困難をきたした。
そこで、平成27年度は、C.japonicaのゲノムを自身で再解読し、良質なゲノム配列を得て、遺伝子機能の解析に使用できるようにすることを試みた。
その結果、10世代系統選抜により作出した遺伝的に均一なC. japonicaの複数株を得た。その中から宿主探索等の形質を維持した株を選び出し、現在全ゲノム情報を解析中である。全ゲノム情報は解析中であるが、ミトコンドリアのゲノム配列解析については完了し、同属の線虫と遺伝子配置の類似性が高いことが明らかとなった。
本課題は、本年度で終了となったが、線虫の重力認識および長期生存の「機構の解明」という点では未完である。しかし、本研究でこれまで得られた情報をもとに、完成は可能であると考えられる。また、本研究の進行時に、本研究の目的とは異なるものの、学術的に重要な線虫Caenorhabditis sp.34が発見された。この線虫はC. elegansの最近縁種にも関わらず、イチジクで増殖し、イチジクコバチに運ばれるというC. japonicaと類似した生活史を持っていることが明らかとなった。現在この線虫の全ゲノム情報の取得および生態の詳細を調査中である。今後は本研究課題を発展、完成させるとともに、発見された線虫の解析を進めていく。
