Research Abstract

生理的な細胞増殖と細胞接着のシグナル伝達機構の解明は、がんの生物学的特性である無制限な増殖能や浸潤・転移能を分子レベルで理解する上で必須であると考えられる。このような観点から、私共は細胞の増殖と接着の制御機構における蛋白質チロシンリン酸化シグナルの生理的役割とがん化や転移の病態との関連につき研究を行っており、本年度は、新しい細胞間シグナル伝達系であるCD47-SHPS-1系に関連する分子の生理的機能と作用機構を中心に研究を行い、以下の結果を得た。
(1)糖鎖認識型ユビキチンリガーゼであるNFB42はSHPS-1の細胞外領域に結合し、SHPS-1を基質としてユビキンチン化し、SHPS-1の細胞表面への発現を制御することを明らかにした。
(2)赤血球上のCD47はマクロファージ上のSHPS-1と相互作用して、赤血球貪食を負に制御する。一方、SHPS-1の類縁分子であるSIRPβはMAPキナーゼ系を介してマクロファージ貪食能を正に制御していることを明らかにした。
(3)神経芽腫由来細胞において、CD47の強制発現により突起伸長が刺激された。CD47の強制発現による突起伸長にはRacが、SHPS-1-Fcによるfilopodia形成にはCdc42が関与すること、また前者の効果にはintegrinの関与が示唆された。
上記の如く、当初の研究目的を充分に達成することが出来た。
