Research Abstract

生体内各器官の形成と維持においては、それらを構成する種々の細胞群の増殖と分化がサイトカインなど細胞外来性のシグナルにより精妙に制御されることが必要であり、その破綻が発癌に至ると考えられる。また、癌の再発や転移に癌幹細胞が関与していることが示唆されていることから、癌幹細胞の特性を知り、その維持機構や増殖・分化制御機構を解明することは癌幹細胞をターゲットとした治療法の開発に重要であると考えられる。本研究は、細胞増殖を正に制御するサイトカインシグナルと負に制御するシグナルの相互作用に焦点をあてた癌細胞の増殖制御機構の解明を目指すとともに、癌細胞株における癌幹細胞の同定とその悪性度への関与の解析を目的とした研究も実施した。前者のプロジェクトについては特に、SCF、bFGFおよびOSMの3つのサイトカインで血球が出現する胎生期マウス造血組織であるAGM領域のin vitro培養系を用いて、細胞増殖を負に制御するしくみに取り組んだ。AGM領域で起こる血球細胞の輩出機構の詳細を明らかにするために、OSMによるシグナルの下流で働き、AGM領域から輩出される血球細胞の増殖に関与する転写因子STAT3をリン酸化するHIPK2の血球細胞分化に対する影響を解析した。AGM領域についE10.5胚の大動脈とその周辺領域におけるHIPK2の発現を認めた上で、野生型HIPK2をAGM分散培養系に強制発現させたところ、未分化血球マーカー陽性細胞の輩出が減少することを確認した。この結果から、HIPK2は血球細胞の輩出を負に制御していることが示唆された。さらにHIPK2の各ドメインの機能を欠失した変異体を用いて解析したところ、血球細胞輩出の抑制には核移行シグナルを含む配列とキナーゼドメインが重要であることがわかった。また後者のプロジェクトについては、グリオーマ細胞株C6における癌幹細胞を、Hoechst33342 dyeでの染色性に基づいて同定し、その悪性度への関与を明らかにすることができた。
