Research Abstract

がん患者において腫瘍細胞に対する有効な獲得免疫反応の誘導に好適な自然免疫反応の促進と、それらを仲介する樹状細胞の機能に着目して研究を進めた。
樹状細胞が分泌するサイトカインの一つでありIL-12と構造上・機能上数多くの点で関連を持つIL-23について、マウスの前脛骨筋肉内に発現プラスミドをin vivo electoporation法で遺伝子導入して強制発現させることにより、IL-23蛋白を継続的に全身投与したのと同じ状況を構築し、その抗腫瘍効果ならびに免疫学的機序について検討した。その結果、IL-23は有意な抗腫瘍効果を有しており、その効果はIL-12と同等のものであった。しかし、IL-12治療の際に見られるような血清中INF-gammaの上昇は見られず、抗CD4抗体あるいは抗CD8抗体を用いた場合、IL-23の抗腫瘍効果が完全に消失することが判明した。これに比して、抗AsialoGM1では、抗腫瘍効果は有意に減弱するものの完全消失は見られなかった。よって、IL-23による抗腫瘍効果の機序はIL-12のそれとは大きく異なることが示唆された。
また、上記のIL-23の検討に用いられたのと同じIVE法を用いて、骨髄中の幹・前駆細胞がレセプターを持つサイトカインであるFlt3-Lの抗腫瘍効果について検討した。Flt3-ILの大量投与単独では有意な腫瘍縮小効果は得られなかったが、肝臓や脾臓を始めとする末梢臓器に大量の樹状細胞を動員され、担癌マウスにおいては皮下腫瘍部にも樹状細胞が動員され集籏することが判明し、他の免疫学的刺激を加えれば有効な癌免疫療法を構築できる可能性が示された。
以上の結果より、これらの知見を総合的に理解し、かつ、更に検討して上で臨床応用すれば、新規がん免疫療法の開発が可能となると考えられた。
