Research Abstract

がんの遺伝子診断を発展させるためには、診断の標的となるがん関連遺伝子を新規に同定すること、並びに有用な診断法を確立することが重要である。これまでに、非小細胞肺がん抑制蛋白質TSLC1と、その分子経路を明らかにしてきた。
1.TSLC1は免疫グロブリン・スーパーファミリー接着分子をコードし、アクチン結合性蛋白質DAL-1/4.1B、並びに広義の細胞極性に関わる蛋白質MPP3と結合する。そこで第一にTSLC1,DAL-1のがんの診断・治療の標的分子としての意義を検討した。そしてDAL-1のメチル化による不活化が原発性非小細胞肺がん103例中59例(57%)で認められること、特に腺がんでは臨床病期の進行に伴って有意にメチル化の頻度が上昇し、また腫瘍のメチル化が患者の全生存率、無再発生存率と有意な逆相関を示すことを見出し、DAL-1のメチル化が肺腺がんの予後因子となり得ることを明らかにした。
2.一方、共同研究により、リンパ球では全く発現しないTSLC1が、原発性ATL芽球の全例で異所性発現を示すことを見出し、ATLに特異的な診断、治療の分子標的となり得ることを示した
3.アレル特異的遺伝子発現の高度定量的検出法であるRNA Difference Plot (RDP)法を確立し、遺伝性腫瘍の同一家系内の保因者間で、変異アレルの相対発現量が著しい不均衡を示し、個人の表現型を修飾する可能性があることを示した。
以上の解析により、TSLC1が上皮、神経由来の腫瘍の大部分で、特にその進展に伴い不活化し、がん抑制遺伝子として機能することが、さらに確認された。一方、TSLC1を介する細胞接着が、ATL細胞では組織浸潤や腫瘤形成といったATLに特徴的な病態の形成に関与する可能性が示唆される。ATLではTSLC1が治療標的分子としても働く可能性も示唆される。
