Research Abstract

【目的】Reg(Regenerating gene)遺伝子は144アミノ酸の分泌蛋白質、Reg蛋白質をコードし、Reg蛋白質は膵β細胞のReg受容体にオートクリン・パラクリンに作用して再生増殖を引き起こす。Reg蛋白質を糖尿病モデル動物に投与すると、膵β細胞が再生増殖し、糖尿病が治癒することが示され、糖尿病の再生治療への応用も期待されている。本研究では、Reg蛋白質による細胞周期調節を検討した。【方法・結果】膵β細胞株のRINm5F細胞の培地にReg蛋白質を添加、刺激したところ、(1)サイクリンD1量、サイクリンD1 mRNAの増加、(2)サイクリンD1プロモーターの活性化が認められた。(3)サイクリンD1プロモーターの活性化は上流-60までを含む場合は保たれたが、-57〜-52の転写因子ATF-2結合配列に変異を導入すると、消失した。(4)ドミナントネガティブ型ATF-2の発現によりReg蛋白質によるサイクリンD1プロモーターの活性化が抑制された。(5)PI3キナーゼ阻害剤によりReg蛋白質によるATF-2のリン酸化とサイクリンD1プロモーターの活性化が抑制された。(6)抗PI3キナーゼ抗体による免疫沈降物は用量依存的にATF-2をリン酸化した。(7)Reg遺伝子欠失(KO)マウスを作製した。KOマウスラ島では、リン酸化ATF-2、サイクリンD1、リン酸化Rb、BrdU取り込み、がいずれも著明に減少していた。【考察・結論】Reg蛋白質刺激により膵β細胞でPI3キナーゼの活性化が起こり、PI3キナーゼによりATF-2がリン酸化される。リン酸化ATF-2はサイクリンD1プロモーターに結合してサイクリンD1の転写を活性化する。この結果サイクリンD1量が増加し、サイクリン依存性リン酸化酵素の活性化が起こり、膵β細胞の細胞周期がG1からSへと移行すると考えられた。
