Research Abstract

癌の本質は、「足場非依存増殖」にある。本研究は、v-Src癌化細胞を用いて、足場非依存増殖センサーであるSHPS-1と変異型caveolin-1の機能、足場非依存増殖制御系を明らかにするものである。近年、細胞増殖機構の研究は長足の進歩を見せたが、癌の本質である「足場非依存増殖」の機構は、殆ど明らかでない。何故か。それは、増殖機構の研究が、正常細胞のそれに収束してしまった事、そして「足場依存増殖」を制御するセンサーが同定されなかった事による。我々は独自の実験系で、2種の足場非依存増殖制御系を初めて発見した。即ち、v-Src癌化細胞より足場非依存増殖を選択的に阻害するSHPS-1を、ヒト乳癌組織より足場非依存増殖と浸潤を同時に活性化する変異型caveolin-1を発見した。本年は、足場依存シグナルの詳細を明らかにする事を目指し、SR3Y1のフィブロネクチン状への接着の、SHPS-1の有無による変化を解析した。まず,SHPS-1発現をTet inducible systemによって可逆的に制御できるSR3Y1株を樹立した。この細胞を用いて、接着の変化を解析したところ、SHPS-1の発現により劇的に接着が回復する事、この時細胞はruffling構造を回復する事、focal contact様構造が回復する事が明らかになった。更に我々は、SHPS-1依存シグナルを探索し、炎症性サイトカインのシグナルにはSHPS-1,SHP-2が重要な役割を担う事を発見した。現在更に詳細な解析を進めている。
