Research Abstract

Gemininは、mitosisの間にだけに分解される分子を網羅的にスクリーングすることによつて発見された分子である。G1期には全く存在しないが、S期からG2期、M期の開始にかけて蓄積し、有糸分裂の中期(metaphase)から後期(anaphase)への移行期に急速に消失する。このタンパク量の急激な変化はAPC/Cによるユビキチン化によって分解を受けるためである。また、Gemininを過剰発現するとDNA複製が抑制されることから、DNA複製のライセンシングに関与する分子であると考えられてきた。このように細胞周期の進行に強く関わっていると考えられる分子Gemininのノックアウトマウスを作製することによって、そのin vivoでの生物学的役割を検討することが本研究の目的である。
Geminin遺伝子をクローニングし遺伝子構造を明らかにした後、Cdt1結合領域をコードするエクソンをネマイシン耐性遺伝子に置換するターゲティングベクターを作製し、定法に則りノックアウトマウスを作製したところ、胎生早期に致死であることが確認された。現在までの解析では、ノックアウト胚は胎生7.5日には観察されていない。このような胎生初期の死亡からも細胞の正常な分裂に必須の分子であることが予想される。
一方、2004年には、Gemininは、Polycomb遺伝子との相互作用によって、Hox遺伝子を制御することが報告された。今後は、分化におけるHox遺伝子との相互作用も検討したいと考えている。
