Research Abstract

骨格の発生過程においては複雑な骨および軟骨のそれぞれの組織のシグナルのクロストークにより最終的な形が作られる事が推察されている。特に軟骨膜は骨基質そのものの上にありながらPTHrP受容体を発現する一方でGliなどの転写因子を発現し、骨格の形の決定ならびに適切な成長の調節を行っていると推察されている。しかしながら軟骨膜の骨格形成における役割は未だ十分には明らかにされていない。そこでトリの受精後7日例の脛骨を12日例のトリ漿尿膜上おいた器官培養(CAM培養)を行った。この漿尿膜培養系(in ovo)では血管の侵入を伴った軟骨膜からの膜性骨の形成が可能であり、これにより生理的な状況における骨膜と軟骨の相互作用を検討した。対照としては血管形成が生じない静置培養を行った通常のin vitroの器官培養系を用いた。軟骨膜を剥離したin vitroの器官培養系では既に報告のあったように軟骨膜の非存在により長軸方向への軟骨の伸長が増加した。一方、in ovoにおいて骨形成を十分に補償した漿尿膜上培養の系においてはこのような骨膜の除去による増殖の亢進は抑止された。以上のことから正常に発育する軟骨膜のシグナルがin vitroでは骨の長軸への成長を抑止する一方でin ovoにおいては血管侵入の結果骨形成が十分に補償されることにより軟骨膜の存在下でも長軸方向への骨の成長への抑止は阻害する因子の存在する可能性が示唆された。このような骨の形成は軟骨膜の非存在下でのアグリカン遺伝子の低下を伴っているが,in vitroの器官培養の如く軟骨膜の存在下でのアポトーシスの亢進はin ovoにおいては見られなかった。以上の結果、軟骨、骨の発生における長管骨の形態形成の上で軟骨膜と軟骨、骨とのシグナル相互作用が存在することが明らかとなり、成長の抑制因子は血管の侵入によって軟骨膜の非存在下でも骨形成の伸長する条件では、除去されることから骨芽細胞自身のこの制御過程における関与が推察された。
