Research Abstract

精子幹細胞は精子形成の源にある幹細胞である。精子幹細胞の細胞の直接の前駆細胞である始原生殖御はLeukemia inhibitory factor (LIF),basic fibroblast growth factor(bFGF),Steel factor (S1)の存在下で培養するとEmbryonic stem (ES)細胞様の細胞に変化し、Embryonic germ (EG)細胞として多分化能を獲得することが知られている。我々は精子幹細胞も培養条件においては可塑性を示すのではないかと考え、精子幹細胞の培養実験を行った。
Epidermal growth factor,bFGF,Glial cell-derived neurotrophic factor,LIFの添加により、胎児線維芽細胞上で培養を行い、マウス精子幹細胞を5ヶ月以上に期間にわたり増幅を行うことに成功した。増幅された幹細胞は不妊マウスの精細管内に移植することで、正常な精子・子孫を作ることができた。これまでに生殖細胞を増幅する方法としてはES細胞とEG細胞の二つが存在するが、この両者と区別するために我々はこの培養細胞にGermline stem(GS)細胞と名付けた。この方法は精子幹細胞における遺伝子操作の基盤技術となるものである。
我々はこのGS細胞の分化可塑性を調べるために、成体マウスへの静脈注射実験、胚盤胞への、移植実験を行ったが、現在までのところ可塑性を認めるには至っていない。現在我々は試験管内において培養条件を更に変化させることで多分化能を持つか否かを検討している。
