Research Abstract

葉の向背軸形成に関わる遺伝子として,AUXIN RESPONSE FACTOR (ARF)遺伝子の発現をteb変異株について調べたところ,ETTIN (ETT)/ARF3やARF4は,teb変異株ではmRNA存在量が上昇し,tebatrの二重変異株では,さらにその量が上昇していた。ETT/ARF3やARF4の発現上昇はtsk変異株でも確認された。また,葉の向背軸形成に関連する他の遺伝子との関わりをみるために,tebと各遺伝子変異株との二重変異株の作製し,その影響を調べた。asymmetric leaves2(as2)teb二重変異株は,teb単独変異で見られる葉の向背軸形成に関わる表現型を元進し,as2,argonaute 7(ago7)の二重変異株について報告された(Xu, L.et al.2006),棒状やラッパ状の葉の形成が観察された。しかし,ago7,teb二重変異株は,葉の向背軸形成に関わる表現型を若干亢進するに留まった。これらのことから,TEBはAGO7,RDR6,SGS3経路によるtrans-acting siRNAとは別経路でETT, ARF4の発現制御に関与するのでは無いかと考えられた。しかし,葉の向背軸形成に関わる遺伝子でも,向軸側形成に関わる遺伝子であるphabulosaやrevoluta変異はteb変異株の形態に明白な影響を与えなかった。
