Research Abstract

我々は、シロイヌナズナのASYMMETRIC LEAVES1(AS1)とASYMMETRIC LEAVES2(AS2)遺伝子を基軸として、葉の発生分化の分子的仕組みを研究している。本年度は、(1)as1,as2変異体におけるClass 1 KNOX遺伝子の転写上昇とサイトカイニン合成酵素遺伝子の発現との関係を明らかにすること、(2)as1,as2変異と遺伝的に相互作用する遺伝子を同定すること、(3)AS1とAS2タンパク質の細胞核内の局在を調べること、(4)Class 1 KNOX以外に、as1,as2変異体で転写レベルが変動している遺伝子を同定すること、(5)根におけるAS2遺伝子の役割を解明し、AS2,AS1の葉の形態形成における包括的な理解をめざした。本年度は以下のようなことを明らかにした。
(1)Class 1 KNOX遺伝子とサイトカンイン合成酵素遺伝子の発現:すでに、STMやBP/KNAT1を過剰発現するとAtIPT7 mRNAの蓄積レベルが上昇することが報告されている。しかし、野生型、bp/knat1,2,6変異株のいずれの株においてもmRNAの変動は見られなかった。
(2)as1,as2変異と遺伝的に相互作用する遺伝子の解析:as1とas2の変異を亢進する変異遺伝子を同定したところ、siRNAの生成に関わる因子(RDR6)、クロマチン修飾に関わるタンパク質(HDACなど)の遺伝子であった。
(3)核内局在:AS1,AS2ともに核小体に隣接する構造小体に局在した。
(4)as1,as2変異体で転写レベルが変動している遺伝子の同定:アレーの解析により、新たにTAS3,ARF3/ETT, ARF4などが変化していることがわかった。
(5)根におけるAS2遺伝子の役割:根ではAS2はコルメラ細胞でのみ発現しており、その細胞分化に正に作用している可能性がある。
