Research Abstract

葉や花弁などの側生器官の背腹を決定する場合には分裂組織と器官原基との相対位置が重要な要因となる。本研究では、シロイヌナズナの突然変異体を用いて、器官の基本構造決定の遣伝的な制御機構を解析した。
(1)側生器官の背側マーカー遺伝子PHBの発現領域を解析した。PHBの発現領城は、PHBpromoter::YFPの発現と、GFP遺伝子内部にmicroRNA165/166による認識・切断配列を挿入して35Sプロモーターにつないだ場合の発現を比較した。PHBp::YFPは茎頂分裂組織のほぼ全域で発現しているが、35S::GFP-miRsiteの発現は原基が盛り上がる前の葉原基の予定領域では低下しており、その場所で腹側マーカー遺伝子FILが発現していた。この結果は、葉原基形成の極初期に表側と裏側領域が区別されて特異的遺伝子の発現領域の境界が決定されており、microRNAが活性を持っていることを示している。また、後期球状胚においてPHBp::YFPが胚の上部ほぼ全域で発現するのに対し、35S::GFP-miRsiteはV型の発現パターンを示したことから、胚発生の初期においてもmicroRNAが機能発現すると考えられる。
(2)FIL遺伝子の発現パターンが変化する突然変異体の解析を進めた。#2-07-4変異株ではFILの発現領域が広くなるが、維管束組織の位置はほぼ不変であった。この結果は、葉断面における維管束の位置はFIL-PHB発現領域の決定機構とは独立に決められることを示唆する。
(3)葉の周縁部の細胞列形成に必要でこの細胞列で特異的に発現するPRESSED FLOWER遺伝子および花弁原基でのみ特異的に発現領域するRABBIT EARS遺伝子の特異的発現機構を調べるために、プロモーター領域を細分して機能解析をおこない、特異的発現を促す領域を約200塩基対に縮めた。
