Research Abstract

1)SAM形成に関わるるCUC1(CUP-SHAPED-COTYLEDON1),CUC2遺伝子の下流候補遺伝子をマイクロアレイ解析で多数同定し、それらに対して心臓型胚における発現をin situ hybridizationで調べた。その結果、これらの多くの遺伝子が、CUCが発現する所で発現しており、cuc1cuc2二重変異株胚の対応する領域で発現が見られなかった。ただ、これらの中には、野生株でもcuclcuc2でも胚の中で他の領域で特異的な発現を示す遺伝子が幾つかあった。そして、それらの遺伝子の一つであるLAS(LATERAL SUPPRESOR)をCUC2のプロモーターで発現させた所、cuc2cuc3の地上部が示す異常を抑圧した。これは、LASがCUCの下流でSAM形成に関わる事を示唆する。
2)側根基部の細胞分裂を制御するPUCHI遺伝子の変異株は、地上部において花茎の基部に一対の突起物を生じ、その間に構造物が出来る。分子遺伝学的な解析により、この突起物は托葉である事が強く示唆された。また、花芽におけるPUCHIの発現を調べた所、花芽初期に一過的に花芽の上側に発現する事が判った。これらの結果と、野生株と変異株の初期の花芽の形態学的観察の結果から、PUCHIはシロイヌナズナの野生株において花芽分裂組織が形成される時、その根元で包葉が発達するのを阻害している可能性が示唆された。
3)シロイヌナズナの胚頂端部において、オーキシンの細胞外排出タンパク質PIN FORMED1(PIN1)の分布が2枚の線対称な子葉構築に関わり、PID(PINOID)はPIN1タンパク質の働きを後期心臓型胚期から強める事をこれまでに明らかにした。我々はpidエンハンサーmacchibou(mab)1-4を単離し解析しているが、MAB4はNPH3ファミリーのタンパク質をコードしており、PIN1タンパク質の細胞内局在に関与する事を明らかにした。そして、MAB4タンパク質が主として細胞内の機能未知の小胞に局在し、それらの小胞にPIN1やPIDも共局在する事も明らかにした。
