Research Abstract

本研究は,自然界の太陽光下水環境系での光機能界面モデルとして,光照射下の空気/水溶液界相を設定し,この界相での活性酸素生成の検証及びそのための新たな検出法の開発を目的として行われ、次の成果を得た。
1.水環境系での活性酸素の測定には,その寿命や生成環境を考えるとオンサイト測定法が必要であり,そのための新たな電気化学的検出法を提案した。
(1)H_2O_2分解触媒の存在下ビオロゲンの自己組織化単分子膜(SAM)修飾電極(直径:1.6mm)を用いることにより、0.1μM〜10mMのH_2O_2を検出できることを明らかにした。
(2)直径15μmの金ディスクを300μmの間隔で配列させた"金マイクロデスク配列電極"を用いることにより0.1μM〜100μMのH_2O_2の検出が可能であることを明らかにした。
(3)金電極に構築したシステインSAM上にスーパーオキシドジスムターゼ(SOD)が安定に固定され,その直接電極反応が著しく促進されること,そしてこのSOD修飾電極が"第3世代のスーパーオキシドイオンバイオセンサー"として機能することをはじめて明らかにした。
2.酸素/水溶液(飽和の腐植酸を含む)界面に光照射した場合(Xeランプ,98 mW/cm^2),1.9n mol/hのH_2O_2の生成を確認した。また,光を照射しない場合及び窒素雰囲気下ではH_2O_2は生成しないことがわかった。
