Research Abstract

本年度は、昨年度にサイトカインLIFの下流でES細胞の未分化状態維持に関与している分子(未分化性維持因子)の候補として見い出したβ-cateninとGABPαについて、重点的に解析を行った。β-cateninに関しては、(1)LIF刺激によって核内に存在するβ-cateninの安定性が亢進されること、(2)β-cateninの活性型変異体を発現させることによってES細胞の未分化状態がLIF非存在下でも維持できること、(3)未分化性維持因子であるNanogの発現を促進していること、(4)Nanogの発現調節を行っている未分化性維持因子Oct-3/4と結合することを見い出した。これらの結果から、β-cateninはOct-3/4と結合してNanogの発現を誘導することによってES細胞の未分化性の維持に寄与していることが明らかとなった。
GABPαについては、(1)LIF除去によってES細胞の分化を誘導するとその発現が低下すること、(2)GABPαを過剰発現させたES細胞は分化させてもOct-3/4の発現が維持されること、(3)GABPαの発現をRNAi法によって抑制するとEs細胞がLIF存在下でも分化すること、(4)この時Oct-3/4の発現のリプレッサーであるCdx2やCoup-tf、GCNFの発現が誘導されていることを見い出した。これらの結果から、GABPαはOct-3/4のリプレッサーの発現を抑制することによってOct-3/4の発現を調節することによりES細胞の未分化状態維持に関与していることが明らかとなった。
以上の締果から、ES細胞の未分化性維持においてβ-cateninやGABPαが重要な役割を果たしていることが明らかとなった。
