Research Abstract

これまでに本研究において同定と機能解析を進めてきた遺伝子群について、マウス胎児性ならびに成体性線維芽細胞における多能性誘導能を網羅的に解析した。この結果、Oct3/4,Sox2,Klf4,cMycの4因子の組み合わせにより、これらの線維芽細胞に多能性を誘導することが可能であることを明らかにし、報告した(Takahashi, K. and Yamanaka, S., Cell, 2006)。さらに、これら4因子のうち、マウスES細胞における機能が明らかでなかったSox2とKlf4について、その解析を進めた。まず、Klf4については、これがES細胞に於けるLefty1プロモーターの活性化においてOct3/4およびSox2と協調的に働くことを明らかにした(Nakatake, Y. et al., Mol.Cell.Biol., 2006)。また、Klf4の強制発現が、マウスES細胞のLIF非依存性自己複製を維持しうることも見出している。一方、Sox2については、その機能がマウスES細胞の自己複製に必須であり、Sox2機能喪失は栄養外胚葉への分化を誘導すること、そしてSox2のマウスES細胞における主たる機能はOct3/4の転写活性維持にあることを明らかにした(Masui, S. et al., Nat.Cell.Biol., in revision)。さらに、マイクロアレイ法を用いた解析により、Oct3/4の標的遺伝子候補を網羅的に同定し(Matoba, R et al., PLoS One, 2006)、Klf4ならびにSox2の標的遺伝子についても同様の解析を進めることにより、マウスES細胞に於ける遺伝子発現制御の全体像を明らかにすることを現在試みている。
