Research Abstract

初年度はべん毛モーター・トルク発生器周辺のタンパク質の探索とその構造解析とに全精力を傾けることを目標とした。そのためには、すでに多くのことがわかっているサルモネラ菌だけでなく、他の種の未解決のべん毛モーターの解析も必須である。今年度は淡水性の光合成細菌ロドバクター(Rhodobacter sphaeroides)と下水に棲むコーロバクター(Caulobacter crescentus)のべん毛精製法の確立をめざした。
ロドバクターのべん毛モーターは一方向にしか回転せず、時折ストップ・モードを示すのが特徴である。精製したべん毛基部体はサルモネラ菌のものと比べて酸に弱いが、熱には同じように強いことがわかった。また、フックが通常のように曲がっているのではなく真っ直ぐであった。この直線状フックは多形性のため一過的にそうなっているのではなく、様々なpH条件下でも直線的であることからロドバクター特有の性質と考えられるが、べん毛モーターの一方向性とフックの形態とがどのように関係するのか未解決の問題である。この成果はJ.Bacteriol.誌に投稿し、現在第1段階審査を経て改稿中である。
コーロバクターのべん毛は極単毛で、細胞周期のある段階で菌体から放出される。この積極的べん毛放出にはべん毛基部体の破壊が伴うことが示されていた。これに呼応して、我々が精製したべん毛基部体はサルモネラ菌と大きく異なり、トリプシン分解されやすいことがわかった。しかし、トリプシン分解だけではロッドまで破壊することができず、したがってべん毛放出はできないはずである。現在べん毛破壊の他の条件を探している。この成果の一部は1月の国際学会で発表した。
