Research Abstract |
MINIATURIZATION OF THE SAMPLE PREPARATION PROCESS AND THE SUBSEQUENT CHROMATOGRAPHIC SEPARATION PROCESS HAS BEEN REGARDED AS ONE OF THE KEY TECHNOLOGIES INCLUDING AN EFFECTIVE ON-LINE COUPLING OF THESE TWO MAJOR ANALYTICAL PROCESSES. COMPARED WITH TRADITIONAL SOLVENT-SOLVENT EXTRACTION," THE MINIATURIZED, SAMPLE PREPARATION TECHNIQUES COULD SIGNIFICANTLY REDUCE THE USAGE OF ORGANIC SOLVENTS AS WELL AS OTHER CHEMICALS. DOWNSIZING OF THE CHROMATOGRAPHIC SEPARATION SHOULD BE ACCOMPLISHED AT THE SAME TIME TO MAXIMIZE THE ADVANTAGEOUS FEATURE OF THESE MINIATURIZED SAMPLE PREPARATION METHODS. AS THE MINIATURIZED SAMPLE PREPARATION TECHNIQUE, THE AUTHORS HAVE DEVELOPED A NEW TYPE OF EXTRACTION METHOD, SO-CALLED "FIBER-IN-TUBE" SOLID-PHASE EXTRACTION (FIT-SPE), WHERE SEVERAL HUNDREDS OF FINE FILAMENTS OF SYNTHETIC FIBER WERE PACKED LONGITUDINALLY INTO A CAPILLARY. L COMPLEX SAMPLE MIXTURES SUCH AS ENVIRONMENTAL AND BIOLOGICAL SAMPLES. THE EXTRACTION CONDITIONS, SUCH AS EXTRACTION FLOWRATE AND TIME, TYPE OF FIBROUS MATERIALS USED, WERE SYSTEMATICALLY STUDIED AND ALSO SEVERAL PARAMETERS FOR THE AUTOMATION OF THE SAMPLE PREPARATION PROCESS WERE OPTIMIZED ALONG WITH THE DEVELOPMENT OF COMPUTER-CONTROLLING SYSTEM FOR THE EXTRACTION PROCESS. FIBER-PACKED SEPARATION COLUMNS HAVE BEEN INTRODUCED FOR THE SEPARATION OF THE ABOVE SAMPLES IN LIQUID CHROMATOGRAPHY (LC), CAPILLARY ELECTROCHROMATOGRAPHY (CEC) AND GAS CHROMATOGRAPHY (GC). THE SUCCESSFUL COMBINATION OF THE MINIATURIZED SAMPLE PREPARATION WITH FIBROUS EXTRACTION MEDIUM AND THE MICROSCALE SEPARATION WITH FIBROUS SEPARATION MEDIUM ENABLES THE DEVELOPMENT OF HIGH PERFORMANCE MICROSCALE ANALYTICAL SYSTEM HAVING A HIGHER EFFICIENCY, COST-EFFECTIVE FEATURES AND ENVIRONMENTALLY-SAFELY CHARACTERS.
