| Research Abstract |
The transplantation of encapsulated growth hormone-secreting cells represents one potential means of treating GH deficiency. The present study investigated the ability of encapsulated GH3 cells, which are derived from rat somatotropin secreting pituitary rumor, to supply GH in the long term and to effect functional improvement in the rat models. Following polymer encapsulation, GH3 cells were transplanted into the rats. A suspension of 6x10^6 cells in agarose gel was injected into a Millipore chamber consisting of a Millipore filter ring and two Isopore disc Polycarbonate hydrophilic 0.4 μm Durapore filters, which were attached to the ring with Aron Alpha. This chamber was implanted into Wister rat by making subcuatenous tunnel. The secretion of GH from the encapsulated cells, and functional changes in the host animals were examined 3 months after transplantation. Analysis of transplanted rat showed that serum GH level were elevated 3 month after tranaplantation. However, there is no statistical difference in tail length and the weight between microcapsule transplanted rats and untreated rats. Analysis of retrieved capsules revealed that the GH3 cells survived and continued to release GH even 3 months after transplantation. These results further support the potential use of this approach for the treatment of GH deficiency.
On the other hand, we constructed original microcapsule based on 2-hydroxy-ethyl acrylate(HEA) polymer gel. We could get size fractionation between IgG and pituitary hormone size of dextran by this HEA based polymer.
In clinical research, we found that apparent diffusion coefficient of craniopharyngiomas was higher than that of pituitary adenomas. We found a positive relationship between VEGF expression and hemorrhage in pituitary adenoma. And we reported that assessment of the postoperative endocrinologic status should be delayed at least one month after surgery in acromegalic patients.
