Research Abstract

Recently, scaffold biomaterials have been focused in the tissue engineering field. FGMgCO_3Ap-collagen composite was made by mixing the functionally graded CO_3apatite containing Mg with collagen. Modification of gradational Mg^<2+> ions on the apatite crystals promoted cell adhesion. In addition, as a scaffold material, the FGMgCO_3Ap-collagen composite suggested contributing to bone formation. Mg^<2+> ions may contribute to the bone metabolism of osteoclast and osteoblast action with the integrins at their cell surfaces.
Furthermore, to develop a new biodegradable scaffold biomaterial reinforced with a frame, synthesized CO_3Ap was mixed with a neutralized collagen gel and the CO_3Ap-collagen mixtures were lyophilized into sponges in a HAp frame ring with pores. Matrix substances on the pure collagen sponge samples increased with the culture period and the surface of the sponge was almost covered. A sectional view of hematoxylin-eosin staining confirmed that osteoblast cells had well invaded the CO_3Ap-collagen sponge. When these sponge-frame complexes were implanted beneath the periosteum crane of rats, newly created bone was observed toward the inner core of the complex from the surface of the periosteum cranii. These reinforced CO_3Ap-collagen sponges are expected to be used as hard tissue scaffold biomaterials for therapeutic purposes.
