Research Abstract |
MMP is believed to be involved in invasive and metastatic natures in malignant tumors. MMP activities in canine malignant oronasal tumors were investigated by using gelatin zymography, and expressions of mRNA coding MMP-2, TIMP-2, and MT-MMP were also examined by RT-PCR technique. MMP-2/TIMP2 ratio in each histological types were also evaluated, and relationship between these MMP-activities and clinical courses of the host animals was studied. Furthermore, localization of mRNAs of MMP-2 and MMP-9 in canine mastocytoma were investigated using in situ hybridization as well as these activities in the tumor tissues MMP-2 activity measured by gelatin zymograpy was extensively positive in all the tissues from malignant melanoma and squamous cell carcinoma, on the other hand, the benign tumors of acanthomatous epulis and normal gingival tissues possessed lower levels of MMP-2 activity, although the mPNA levels of these MMP families were not significant among the histopathological types in oronasal tumors. However, MMP-2/TIMP-2 ratio of malignant melanoma and squamous cell carcinoma were significantly higher than those of normal gingival tissues. MMP-2/TIMP-2 ratio of the host animals with metastatic lesion. were higher than those without the lesions In the tissues from cutaneous mastocytoma, MMP-2 and -9 activities were detected in 72.2% and 66.6%, respectively. In situ hybridization revealed that the mRNA of MMP-9 were detected in the endothelial cells in tumor tissues, and the signal showing TIMP-2 were detected in stromal cells Based on these results, MMP-2 were closely related to the invasion and metastasis of the canine malignant oronasal tumors, and MMP-2/TIMP-2 ration was of value in evaluating malignancy of the oronasal tumors in dogs. Also in canine cutaneous mastocytoma, both of MMP-2 and -9 were considered to be concerned with malignant natures of the tumor cells. Further investigations on canine MMP families should be conducted
