| Research Abstract |
1.Ghrelin
Previous investigations on the preproghrelin gene reported three common non-synonymous polymorphisms (Arg51Gln, Leu72Met, and Gln90Leu), but their role in susceptibility to eating disorders has not been studied. The purpose of the present study was to determine if any of the three polymorphisms is associated with EDs by a case control study. In addition, we examined if the polymorphisms have any association with anthropometric measures and psychopathologies relevant to EDs in the non-clinical control samples by a cross sectional study. Two hundred and eighty one EDs including 199 anorexia nervosa(AN) and 82 bulimia nervosa(BN) were recruited from collaborating hospitals. Two hundred seventy two healthy volunteers were recruited from students of an university. All participants were female and Japanese. Genotyping of the polymorphisms were performed and distributions of genotype and allele frequencies were compared between the groups. The body height, weight, body mass index, percentage of body fat and scores of the Eating Disorder Inventory-2(EDI-2) were measured and compared between the genotypes within the control. The allele frequencies of Met72 allele were 0.225 for ANR, 0.182 for AN-BP,0.250 for BN and 0.164 for the control. Met72 allele was significantly more frequent in the BN groups than in the control (genotype-wise : p<0.0001,allele-wise : p=0.012). Within the control group, the Met72 carriers showed higher current body weight and higher scores in "Body Dissatisfaction" subscale of EDI-2. The Gln90 allele was rare and Gln51allele was not observed in both patients and controls. Our findings suggested that Met72 allele of Leu72Met polymorphism of preproghrelin gene is associated with susceptibility to BN, and related to body weight and dissatisfaction to body shape in young females.
2.Others
UCP-2 55A/V, UCP-2 ins/del, UCP-2 -866G/A, UCP-3 -55C/T, BDNF Val66Met, AGRP 706G/A and DRD4-521C/T polymorphisms were analyzed and not associated with AN.
