Research Abstract |
We have investigated the feasibility of gene transduction for braintumors using adenovirus vector. Transduction of the cytosine deaminase gene into tumor cells followed by administration of 5-fluorocytosine was found to have strong antitumor effect against experimental malignant brain tumor in vitro and in vivo. We also found its toxicity and limited antitumor effect on infiltrating tumor cells in normal brain tissue which cause local tumor recurrence. The purpose of this research was to improve antitumor effect on infiltrating tumor cells within normal brain tissue while minimizing neurotoxicity on normal brain cells. We developed new method to attain high local concentration of toxic drug coverted by CD transduced cell. We employed neuronal stem cells and bone marrow stem cells which harbour therapeutic gene and track infiltrating tumor cells. We showed their therapeutic effect in vitro when cultured with glioma cells. Then we investigated tracking of stem cells to infiltrating tumor cell in rat brain tumor model. We are now investigating in vivo therapeutic effect using stern cells harboring therapeutic gene.
