Research Abstract

Basonuclin was originally found in human culture keratinocytes. It has three pairs of zinc fingers, a nuclear localization signal, and a serine stripe. It has been demonstrated to bind to rDNA promoter. It is also a cell-type-specific transcription factor expressed in the highly proliferative cells, such as basal layer of epithelial cells and germ cells. UV is known to the main etiological factor in causing skin cancers. Protein synthesis increases during carcinogenesis. We investigated basonuclin localization in UV-exposed skins. After mice dorsal skins were exposed to UV with Toshiba FL20SE fluorescent sunlamps, the specimens including hyperplasia, solar keratosis, ad squamous carcinoma were obtained.
The freshly isolated tissues were snap-frozen in O.C.T. Compound. Frozen sections were cut at 6 μm and then fixed with acetone-methanol. Staining for basonuclin was carried out using an affinity-purified rabbit polyclonal antibody to full-length human basonuclin. Incubation with biotinylated goat antiserum to rabbit IgG, followed by visualization with streptaviden-Cy3 were each carriedout. DNA was stained with Hoechst 33258.
The basal and immediately supra-basal cells contain basonuclin mainly concentratedin the cytoplasm in normal healthy skin. On the other hand, the basal and immediately supra-basal cells contain basonuclin mainly concentrated in the nucleus in hyperplasia and solar keratosis. Squamous carcinoma also contains basonuclin mainly concentrated in the nucleus.
Therefore, it is speculated that basonuclin is required in active protein synthesis during UV induced carcinogenesis.
