Research Abstract |
I.Expression of Doc-1 protein Expression of Doc-1 protein was investigated in 40 patients with leukoplakia of the oral cavity, primarily using tissue samples of atypical epithelia. 1.Methods 1)Western blotting Collected tissue samples were crushed in RIP buffer and centrifuged. Supernatant was then subjected to SDS-PAGE using polyamide gel. Protein in the gel was transferred to a PVDF membrane and the presence and changes in Doc-1 protein were ascertained using the alkaline phosphatase method. Anti-actin antibody was used as a control sample. 2)Immunohistochemical staining Collected tissue samples were frozen and sliced, and according to the indirect fluorescent antibody method, anti-mouse Doc-1 polyclonal antibody was used as the primary antibody. FITC-labeled antibody was then used as the secondary antibody to detect primary antibody. 2.Results and Discussion -In patients in whom leukoplakia became cancerous during the study, Doc-1 protein was detected in both Experiments 1 and 2. -In patients in whom leukoplakia did not change during the study, Doc-1 protein was detected only in Experiment 2. -In patients in whom dysplasia advanced during the study, no defined tendencies were seen in Experiments 1 and 2. The results of this study suggest that if Doc-1 protein is seen by Western blotting or immunohistochemical staining on initial presentation or during follow-up in patients with leukoplakia of the oral cavity, dysplasia is likely to advance and become cancerous. II.Changes in salivary cytokines in patients with leukoplakia of the oral cavity Levels of salivary IL-6 and -8 were measured in 32 patients with leukoplakia of the oral cavity, and results were compared to levels in healthy individuals. 1.Methods ELISA was performed. 2.Results and Discussion Levels of IL-8 were significantly higher for patients with leukoplakia of the oral cavity than for healthy individuals.
