Research Abstract

Neurons continue to be generated in the adult hippocampus. We examined the cell-cell interaction between proliferating cells and immature neurons in hippocampal neurogenic region. Proliferating cells were detected by immunohistochemistry for a cell cycle marker Ki67, and the fate of the proliferating cells were traced by labeling with bromodeoxyuridine or a retroviral vector carrying an enhanced green fluorescent protein. Neural precursor cells appeared to divide at least 3-4 times and formed compact clusters in the subgranular zone and innermost part of granular layer. A cluster contained Ki67-positive proliferating cells, polysialic acid-neural cell adhesion molecule (PSA-NCAM)- and Hu-expressing immature neurons, or GFAP-positive astrocytic cells. A majority of the newly generated cells remained within the cluster for 3-4 days after the generation and a few newly generated cells did for 7 days or more. This indicates that cell proliferation and neuronal differentiation occur within the clusters and as a result newly generated cells within a cluster vary in nature and developmental stages. Thereafter newly generated cells at advanced stages of development migrated from the cluster mostly tangentially. The migrating cells expressed molecular markers for immature neurons such as PSA and Hu, and had tangentially oriented long processes. One tip of the tangentially oriented processes frequently made contact with some cells of the clusters or penetrated the center of the clusters. We assume that cell-cell contact among newly generated cells at different stages of development, including formation of compact clusters and contact by tangentially oriented long processes is involved in cell proliferation and differentiation of neuronal precursor cells.
