Research Abstract

細胞内寄生菌弱毒株を用いたDNAワクチン運搬システムは高い安定性、安い生産コスト、投与技術の簡便さの点から、家畜の次世代ワクチンとして極めて有望である。本研究の目的は、免疫応答の場であるリンパ節に寄生するロドコッカス・エクイ中等度毒力株をDNAワクチンの免疫系(樹状細胞)への効率的な運搬ベクターとして利用可能であるか否かを検討し、経口ワクチンとして家畜の次世代ワクチンの可能性を探ることである。DNAワクチンを考える上でその候補遺伝子抗原についてペプチドワクチンを用いて、以下の検討を行った。R.equi病原性プラスミド上にコードされたVapAのB細胞エピトープを含むペプチド(N15Y),T細胞エピトープを含むペプチド(V20S)及びそのハイブリッドペプチド(NV)の3種の合成ペプチド並びに組み換えVapD(rVapD)蛋白を用いて免疫し、マウスと馬における抗体応答、並びにマウスにおける感染防御能も検討した。その結果、N15Y及びNV免疫マウスでは、ペプチド単独免疫群に比べ不完全又は完全アジュバントを用いた免疫群で抗体価が著しく上昇し、サブクラス別抗体応答ではIgG1とIgMの抗体価の上昇が認められた。しかし、抗体価の上昇に拘わらず何れのペプチドも感染防御能を賦与することは出来なかった。馬における抗体応答としては、NV抗原免疫馬ではIgGbの抗体価が上昇し、続いてIgGaが上昇した。rVapD免疫馬ではIgGaとIgGbが同時期に上昇したが、IgGaの抗体価の上昇が顕著だった。IgMとIgAは観察期間を通しほぼ一定の低値を示していた。
