Research Abstract

本研究の目的は、細胞膜上にヘム蛋白性酸素感受性イオンチャンネルを持つPC12細胞を用い、その細胞膜画分のヘム蛋白をプロファイリングすることにより、新規酸素感受性分子(酸素センサー)を同定することである。
平成14年度中の研究により、Lithium Dodecyl Sulfate (LDS)を用いた低温下ポリアクリルアミドゲル電気泳動法(LDS-PAGE)と、ヘム分子団のペルオキシダーゼ様活性に基づきヘム蛋白を特異的に検出するルミノール化学発光反応を組み合わせた、高感度ヘム蛋白検出法を確立した。
平成15年度の研究は当初計画通りに遂行され、以下の成果を得た。
1.PC12細胞の粗精製細胞膜画分蛋白を低温下LDS-PAGEにより分離後、ルミノール化学発光反応液中でインキュベートしフルオログラフィーをとることにより、粗精製細胞膜画分に含まれるヘム蛋白のバンドを同定した。さらに、同一試料を並行して泳動したレーンをクマジーブリリアントブルーで染色することによって得られた全蛋白の染色バンドと、フルオログラム上のヘム蛋白のバンドの位置を比較することにより、ヘム蛋白を含むと推定される蛋白のバンドを同定した。
2.1.によって同定された蛋白のバンドを切り出し、そこに含まれる蛋白を、液体クロマトグラフィー・マススペクトロメトリー法によって同定した。その結果、約50種の蛋白が同定された。
3.現在、2.によって同定された蛋白の中から、構造や推定される機能などに基づいてヘム蛋白である可能性の高いものを抽出し、ヘムとの結合性を検討中である。すでに、ソーレーバンド(蛋白とヘムとの結合により見られる波長400nm付近の特異的吸収帯)などにより、ヘムとの安定した結合が確認されている蛋白もある。今後、これらの新規ヘム蛋白の機能の解析を進めていく予定である。
