Research Abstract

1 カルボニル還元酵素による血管内皮細胞増殖因子(VEGF)遺伝子発現とp53タンパク質レベルの変化
マウスカルボニル還元酵素(CR)の発現ベクターをヒト卵巣がん細胞株A2780などにリポフェクトアミンを用いて導入した。トランスフェクトした細胞を10%ウシ胎児血清を含むRPMI1640培地で培養した。細胞数はAlamar Blue法で測定し、細胞形態は位相差顕微鏡で観察した。CR、VEGF、p53の発現をウエスタンブロット法で検討した。CR発現ベクターの導入により卵巣がん細胞でCRの発現を認めた。CR発現に伴い、細胞増殖や細胞形態に明らかな変化を認めなかったが、VEGFの発現は低下した。また、CRを発現した細胞、コントロールの細胞ともにp53タンパク質は検出できなかった。
ヒト卵巣がん細胞の抗がん剤、ドキソルビシンに対する感受性が、CRの発現に伴い変化するか検討した。ドキソルビシンによる卵巣がん細胞増殖抑制の程度は、CR発現の有無で明らかな変化を認めなかった。
2 CRによる発現抑制に係わるVEGF遺伝子のしスエレメントの検討
CRの発現によりVEGFのどのタイプが発現抑制を受けるのか、VEGF-A、B、C、Dの各抗体を用いてウエスタンブロット法で検討した。CRにより制御を受けるのはVEGF-Dと同定された。現在、VEGF-D遺伝子のプロモーター領域のコンストラクトを作製し、A2780細胞にトランスフェクトし、CRによる発現抑制に係わる領域を明らかにする準備を進めている。
