Research Abstract

本研究の目的はアルツハイマー病とその動物モデルにおいて、Aβ沈着をより効果的に減少し、副作用の少ないAβ・DNAワクチンを開発することにあった。第一の目標は、有効な遺伝子ワクチンの創製で、産生されたペプチドが免疫担当細胞に留まると分泌型の二型を作成し、抗Aβ抗体産生の程度をみた。その結果、抗体産生の程度には大きな差はないが、Aβ沈着を軽減する効果は分泌方が有意に強かった。第二は、投与量、投与間隔、投与ルートの検討を行った。この分析では筋注と点鼻投与を比較したが、2週1回の投与では両者に大きな差がなかった。これらの所見はAβ・DNAワクチン療法がヒト・アルツハイマー病の特異免疫療法として有効であることを強く示唆している。今後、DNAワクチンを投与する際の担体を改善して、より効果を挙げるようにする計画である。
