Research Abstract

1.GFPトランスジェニックマウスの歯根膜細胞の分離と培養
GFPトランスジェニックマウスの臼歯を無菌的に抜歯し,歯根膜を有する歯根部をコラーゲンゲル内に包埋し,歯根膜細胞をコラーゲンゲル内に遊走させた。細胞が遊走後,コラゲナーゼで細胞を回収し,プラスチックディッシュ上で培養した。これらの細胞はアルカリフォスファターゼ活性陽性で,歯根膜細胞と考えられた。
現在,これらの細胞を野生型骨再生部へ移植すると、これらの細胞が骨芽細胞へ分化していることが確認でき、本研究における当初の実験系を確立することができた。
2.GFPトランスジェニックマウスの骨髄細胞の分離と歯根膜部への移植
GFPトランスジェニックマウスの骨髄細胞を採取し,プラスチックディッシュ上で培養した。ディッシュに付着した骨髄間質細胞を回収し,野生型マウスの歯根膜部へ移植したが、歯根膜部にGFP陽性細胞は確認出来なかった。これは、歯根膜部への細胞の移植法に問題があると考えられたので、現在移植法を検討中である。
3.現在までの研究で歯根膜細胞がin vivoでも骨芽細胞に分化できることを実証する実験系を確立できたが、歯根膜部以外の間葉系幹細胞の歯根膜細胞への分化能の解析を効率良く推進する実験系の確立が不十分だったので、現在、その手法をさらに検討中である。
4.ヒト歯根膜細胞を利用した実験を十分に推進することができなかった。
