Research Abstract

日本分光製のVCDの整備、測定条件の検討を詳細に行った。特に、本装置は、国内2号機にあたるため、同社の技術者と協力し、検出器などの検討を詳細に行った。特に、本装置の得意端数である高端数領域(4000-1600cm-1)の調整を念入りに行い、標準品を設定することにより、CH領域のVCD特性について検討した。
グルコースを単位とする二糖類11種類[Trehalose(α1-α1),Neotrehalose(α1-β1),Kojibiose(α1-2),Nigerose(α1-3),Maltose(α1-4),Isomaltose(α1-6),Isotrehalose(β1-β1),Sophorose(β1-2),Laminarabiose(β1-3),Cellobiose(β1-4),Gentiobiose(β1-6)]のVCD測定を行った。いずれも、グルコース二糖からなる化合物であるが、その結合位置に違いがある。CH特性吸収領域を詳細に検討した結果、結合位置と結合様式の違いを、この領域のCH-VCD吸収パターンで識別可能であるということを発見した。NMR, MS以外では、この識別は非常に困難であり、今後の展開が期待できる。
単糖の代表例として、グルコースの精密理論計算を行った。すべてのコンフォメーションとその存在確立を明らかにし、VCDの理論計算を行った。特にDFT計算における基底関数を検討し、実測値であるグルコースのVCDスペクトルとの比較により、最適の基底関数を発見した。本応用は、VCDをコンフォメーション解析に利用する興味ある試みである。
