Research Abstract

生物は光を情報へと変換するため、光センサーと呼ばれる一群の蛋白質をもっている。既知の光センサー蛋白質は、いずれも光異性化反応によって光情報変換を開始していた。ところが、最近になってフォトトロピンなどの青色センサーは、発色団として異性化が不可能なフラビン類をもつことが明らかになった。そこで本研究では、フラビンを発色団とする光センサー蛋白質の光反応メカニズムの解明を目的として、フーリエ変換赤外分光法などの分光学的手法を用いた解析を行った。その結果、本年度は以下の研究成果が得られた。
植物の青色センサーであるフォトトロピンのフラビン結合部位LOV2に対して極低温から室温に至る幅広い温度領域で可視および赤外分光測定を行った。可視吸収スペクトル測定の結果から、1つの中間体だけが生成することが明らかになった。この結果は、ロドプシンなどの他の光センサー蛋白質と大きく異なるものである。
また赤外分光測定により、フラビンと共有結合を形成するシステイン側鎖はプロトン化していることがわかった。さらに共有結合形成により、蛋白質の内部結合水の水素結合が切れることを見出した。一方、蛋白質の二次二次構造変化をモニターするアミドの領域では温度依存的なスペクトルが得られた。この結果は、1つの中間体しかもたないフォトトロピンが、ロドプシンなどと同様、段階的な蛋白質の構造変化を示すことを示唆するものである。
