Research Abstract

動物は、環境への適応機構のひとつとして神経系を進化の過程で獲得した。刻々と変化する状況に即して動物は様々な行動を発現し、外部環境の変化に適応している。では、動物は状況に応じてどの様な神経生理機構で適切な行動を発現し、切替えているのか?本研究では、いかに状況に応じた行動が発現されるのかその神経機構を理解しようとしている。
神経の可塑的な性質は行動を切り換える際に必要な生理機構である。この様な神経の可塑的な変化には一酸化窒素(NO)が重要な役割を担っていると考え、節足動物、特にザリガニやコオロギを実験材料として、同定可能な神経回路網においてNOがどの様な修飾効果をもつのか細胞レベルでの詳細な解析を進めるとともに、行動レベルでの効果、役割を解析してきた。その成果として、1.組織化学染色法でNO産生細胞と標的細胞の局在をこれまでに確かめ、さらに、昆虫の脳では常にNOが100-200nM程度放出されていること、刺激により更に放出量が増加することを電極法で計測し示した。2.NOの光学的な計測を行い、中枢神経系の中でどの程度の一酸化窒素が産生されているのか突き止めるため、既存の共焦点レーザー走査顕微鏡を改造するため備品としてレーザー光源ユニットを購入し、NO感受性色素を使って光学的に解析を続けている。3.電気生理学的にNO産生細胞と標的細胞の両方の細胞から活動を計測し、一酸化窒素やcGMPの合成を促進或いは抑制する薬物を灌流投与し、一酸化窒素の修飾効果を解析し、神経系におけるNOの修飾効果を示した。
これまでの成果は国際シンポジウム及び国内学会で発表し、また、その一部は既に国際学会誌等に論文として発表し、残りの成果については現在発表の準備に取りかかっている。
