Research Abstract

細胞は、恒常的に生じる二本鎖DNA切断(DSB)を感知し、細胞周期を積極的に停止することにより損傷修復を行うことが知られている。本研究では、放射線によるDSBに対するカイコ細胞の応答反応を解析し、DSB部位認知の分子機構とこれらの修復機構選択の持つ役割を解明することを目的に研究を行った。特に修復機構については、相同組換え修復(HR)に焦点を絞った。
その結果、カイコ細胞は放射線によるDSBが染色体上に生じた後、G_2/M期に細胞周期を停止し、その後、アポトーシスを誘導することなく回復できるため、放射線に対して強い抵抗性を示すことを明らかにした。
また、カイコ染色体上でのHR測定系を複数構築し、これらの測定系を用いて、カイコ細胞がDSBの存在部位(染色体のDNA上の損傷か否か)を識別し、異なる経路で修復できる能力を有することを碓認した。この際、染色体上のDSBの修復には、HRを使用し、SSA(Single-Strand Annealing)はほとんど利用されないことが示唆された。
さらに、HR修復、細胞周期制御に関わるカイコ遺伝子を多数単離し、その中の一遺伝子がDSBの存在部位の認識に関る新規遺伝子であることを明らかにした。この遺伝子は染色体の構造情報を介して、DSBの存在部位を認識している可能性が示唆された。
