Research Abstract

[プリオン蛋白(PrP)の機能とアポトーシスの抑制に関する解析]
Zn-1型PrP遺伝子欠損マウスおよびRikn型PrP遺伝子欠損マウスおよび野生型マウスより得られた神経細胞初代を用いて銅イオンによって誘導されるアポトーシスについて解析した結果、PrP遺伝子欠損マウス由来神経細胞が有為に高いアポトーシスを起こした。
また、Zn-1型PrP遺伝子欠損マウス、Rikn型PrP遺伝子欠損マウスおよび野生型マウスより得られた脾細胞を用いて無刺激および刺激後の細胞の増殖能について比較検討を行った結果、PMA、イオノマイシン刺激後において野生型マウス由来脾細胞と比較してZn-1型PrP遺伝子欠損マウスおよびRikn型PrP遺伝子欠損マウス由来脾細胞において増殖能が低い結果が得られた。ConA、リポ多糖刺激においては野生型マウス由来脾細胞とPrP遺伝子欠損マウス由来脾細胞間で有為な差は認められなかった。アポトーシス細胞についても解析を行ったが有為な差は認められなかった。
[ウイルス感染時におけるプリオン蛋白の機能解析]
Zn-1型、Rikn型PrP遺伝子欠損マウスおよび野生型マウスを用いて、脳心筋炎ウイルス-B株感染について解析した結果、アポトーシス細胞の出現頻度においてPrP遺伝子欠損マウスのほうが、野生型マウスと比較して高頻度であった。
