Research Abstract

胎盤機能不全あるいは子宮内低栄養による子宮内胎児発育遅延(IUGR)の胎児予後を改善する目的で、昨年度までに胎盤形成過程を検討するための移植実験系(項目1)や胎児発育遅延のマウスモデル(項目2)を作成してきた。その成果を踏まえ、本年度はさらに下記の検討を行った。
1.胎盤の形成過程の解明と機能再生をめざした骨髄由来細胞の移植実験系の解析
a.遺伝子操作により全身の細胞を蛍光色素(GFP)標識されたマウス(GFP-Tgマウス)の雌にnon-Tgマウスを交配し、妊娠18.5日目にnon-Tg胎仔の胎盤を採取した。この胎盤の連続切片のほぼ100%に母獣血液細胞より大型のGFP陽性細胞を検出しその内約2%はCD34陽性であった。
b.GFP Tgマウスの大腿骨および頚骨から骨髄由来細胞を調整し、これをnon-Tgマウス(妊娠4.5および6.5日)の尾静脈より注入した。妊娠18.5日に胎盤内への生着の有無を検討したところ、胎盤(n=3)の連続切片の11-18%において1-5個のGFP陽性細胞を検出した。
以上より、母獣骨髄由来細胞が胎盤内に生着し、胎盤形成に何らかの関与をし得る可能性が示された。
2.妊娠マウスの摂食制限による胎仔発育遅延モデルの解析
IUGR胎児では出生後良好な発育をし、いわゆるキャッチアップした児においても成人期に肥満、高血圧、心血管障害、糖尿病など、各種の生活習慣病が高率に発症することが報告されつつある。本研究では子宮内発育遅延モデル動物において、成長後の肥満発症に及ぼす因子を検索した。
a.マウス妊娠母獣の摂食制限により胎仔発育の遅延を認めること(IUGR群)、出生仔の新生仔期に一過性の高レプチン血症を呈することが明らかとなった。
b.上記の出生仔マウスに生後8週齢以降に高脂肪食を付加すると、対照群でも肥満を生じたが、IUGR群ではさらに高度の肥満を呈した。対照群のマウスでは高脂肪食を負荷したときに酸素消費量が増加し、体温の上昇(いわゆるdiet-induced thermogenesis ; DIT)が見られたが(n=8)、IUGR群ではDITが見られなかった。
c.妊娠中に母獣の摂食制限を行わなかった仔に対して新生仔早期にレプチンを投与すると(新生仔期レプチン群)、生後8週齢以降の高脂肪食付加により新生仔期レプチン群では高度の肥満を認めた。
以上から、IUGR児の出生後の一過性高レプチン血症が、成長後の肥満に関与する可能性が示唆された。
