Research Abstract

(1)イネのUVB感受性とCPD光回復酵素遺伝子の構造的変異、酵素特性について
UVB感受性のことなる日本型イネ17品種を材料に、UVB感受性とCPD光回復酵素遺伝子の配列、およびその酵素活性を比較した結果、(1)CPD光回復酵素の遺伝子型は126番目がGln(ササ型)とArg(農林型)の2つの型(他のアミノ酸配列は同じ)に分けられ、ササ型は酵素活性が高く、一方農林型の活性はササ型と比較して低いこと、(2)ササ型の遺伝子を有する品種はUVB抵抗性が高いことがわかり、少なくとも1塩基の変異(377番目)による126番目のアミノ酸変異が、日本型イネ品種のUVB抵抗性を決定することを明らかにした(Teranishi et al.2004)。また、UVB感受性の異なるインド型品種を材料に同様の研究を展開した結果、(1)日本型品種よりもさらに感受性を示す品種は、126番目がArgで、かつ296番目がHisであること、(2)この2つの変異は、基質であるCPDへの結合能低下させることを見出した。さらにUVB感受性の異なる日本型、インド型の後代世代を用いたQTL解析から、これらの変異がUVB感受性を決定する主要因であることを実証した(Hidema et al.2005)。
(2)光回復酵素遺伝子組換え体イネを用いた、光回復酵素とUVB感受性に関する研究
イネ・ササニシキの光回復酵素遺伝子をササニシキ、および農林1号に1コピー導入した形質転換体、および数コピー導入した過剰発現組換え体、ならびにアンチセンスイネを各々数系統作成し、T2、T3種子を獲得した。現在、これら組換え体のUVB感受性試験、発現量・酵素活性解析を行っている。
